- PMID 41857429 (2026, Nature metabolism) — Python metabolomics uncovers a conserved postprandial metabolite and gut-brain feeding pathway.. Abstract (opening): Most mammals consume small and frequent meals. By contrast, pythons are ambush predators that exhibit extreme feeding and fasting patterns and provide a unique model for uncovering molecular mediators of the postprandial response<sup>1-3</sup>. Using untargeted metabolomics, we show that circulating levels of the metabolite para-tyramine-O-sulphate (pTOS) are increased more than 1,000-fold in pythons after a single meal. In pythons, pTOS production occurs in a microbiome-dependent manner via sequential decarboxylation and sulphation of dietary tyrosine. In both pythons and mice, pTOS administration activates a neural population in the ventromedial hypothalamus (VMH). In mice, these VMH neurons are required for the anorexigenic effects of pTOS. Chronic administration of pTOS to diet-induced obese male mice suppresses food intake and body weight. pTOS is also present in human blood, where its levels are increased after a meal. Together, these data uncover a conserved postprandial anorexigenic metabolite that links nutrient intake to energy balance.

Abstract (opening): Modern zoos operate at the intersection of conservation responsibility, ethical accountability and scientific advancement, while facing increasing public scrutiny and rising expectations for transparency and measurable impact. This scrutiny often focuses on animal welfare and the justification for maintaining wild animals in captive environments. Consequently, there is a growing need for zoo-based research to move beyond often anecdotal practice toward robust, evidence-informed decision-making. Evidence refers to systematically collected data and critically evaluated research used to inform management strategies that promote the zoo's key aims. This article synthesises key areas in which contemporary zoo operations rely on limited or untested evidence and identifies priority research questions critical to the future legitimacy and effectiveness of zoos. The paper explores several interlinked themes: the welfare implications of reproduction and breeding management; the role of social bonds in welfare, stress resilience, and translocation outcomes; the underrepresentation of nutrition within animal welfare research; the impacts of human-animal interactions involving both staff and visitors; the maintenance of species-specific behaviours and behavioural diversity in ex situ managed populations; and the relationship between animal welfare, visitor experience, and a zoo's social licence to operate. These themes were identified through persistent assumptions in zoo practice, gaps in the empirical literature, and increasing societal expectations for transparency, ethical justification, and measurable outcomes. Across these areas, the paper highlights a recurring challenge: management practices are frequently justified on theoretical or historical grounds rather than validated, applied outcomes, and conclusions from single studies are often overinterpreted despite limited replication or contextual relevance. Our article therefore emphasises the importance of defining credible evidence, critically appraising published research, and adopting replicated, collaborative multi-institutional and mixed-methods approaches. By outlining these research priorities, this paper provides a framework for advancing zoo-based science that can directly inform management decisions, improve animal welfare outcomes, strengthen conservation impact, build public trust and thus future-proof the zoo industry. Ultimately, we argue that the future success of zoos depends on aligning ethical practice, rigorous research, and meaningful visitor engagement through transparent, evidence-driven approaches.

- PMID 42071908 (2026, Animals : an open access journal from MDPI) — Composition and Legal Aspects of Reptiles and Amphibians Displayed at an Exotic Pet Fair in Warsaw (Poland).. Abstract (opening): The global exotic pet trade has expanded in recent decades, raising concerns related to animal welfare, biodiversity conservation, and compliance with international regulations. Reptiles and amphibians constitute a major component of this trade, yet information on species availability and trade practices at exotic pet fairs remains limited. The primary aim of this study was to identify the reptile and amphibian species offered for sale at an exotic pet fair in Warsaw, Poland. Secondary objectives were to assess the declared origin of the animals and the availability of information regarding their legal and conservation status. Photographic documentation of all exhibition tables was used to record species identity, number of individuals, and labeling practices. In total, 818 individuals representing 74 species from 31 families were recorded. Reptiles, particularly squamates, dominated the assemblage, while amphibians accounted for a smaller proportion of the animals offered. More than half of the individuals were labeled as captive-bred, whereas only a small fraction were identified as imported or wild-caught; however, information on origin was absent for nearly half of the animals. Over 50% of the recorded species were listed in Appendix II of the Convention on International Trade in Endangered Species of Wild Fauna and Flora, yet no visible information on legal or conservation status was provided at the point of sale. These findings indicate that inconsistent labeling limits transparency and informed decision-making by buyers.

- PMID 41123193 (2025, Journal of the American Chemical Society) — Uric Acid Monohydrate Nanocrystals: An Adaptable Platform for Nitrogen and Salt Management in Reptiles.. Abstract (opening): Both avian and nonavian reptiles excrete excess nitrogen in solid form─colloquially termed "urates"─as an evolutionary adaptation that aids in water conservation. Yet, there are many open questions regarding the composition, structure, and assembly of these biogenic materials. Here, analyses of urate excretions from ball python (<i>Python regius</i>) and 20 other reptile species reveal a clever and highly adaptable system employed to handle both nitrogenous waste and salts. Primitive species excrete urates consisting of 1-10 μm microspheres of turbostratic uric acid monohydrate (UAM) nanocrystals. The nanocrystals' high surface area and ionizable nature provides a platform to coeliminate substoichiometric concentrations of various salts through surface-ion pairing. In contrast, the granular urates produced by species from more advanced snake lineages are phase mixtures consisting of predominantly ammonium urate hydrate (AUH) and smaller amounts of other crystalline forms. Identification of microspheres as a minor but highly soluble component of these excretions suggests their likely role as reactive precursors to AUH, a hypothesis supported by in vitro experiments. Importantly, this points to a previously unrecognized physiologic function of uric acid, namely the ability to sequester ammonia by transforming it into a solid. The potential implications of this function in other species are discussed.

- PMID 40804977 (2025, Animals : an open access journal from MDPI) — Identifying Infectious Agents in Snakes (Boidae and Pythonidae) with and Without Respiratory Disease.. Abstract (opening): Mycoplasmas are known respiratory pathogens in tortoises, but few studies exist in snakes. To better understand the correlation with clinical signs and co-infections, samples from mycoplasma-positive snakes with and without clinical respiratory disease were analyzed. Oral swabs from 15 snakes (pythons <i>n</i> = 12, boas <i>n</i> = 3) were examined using polymerase chain reaction (PCR) and third-generation sequencing (TGS). Additionally, mycoplasma isolation assays were performed. Pathogens detected by PCR included Mycoplasmas (15/15, 100%), serpentoviruses (9/15, 60%), and <i>Chlamydia</i> sp. (2/15, 13%); those detected by TGS included Mycoplasmas (14/15, 93%), serpentoviruses (10/15, 67%), <i>Chlamydia</i> sp. (1/15, 7%), and 15 different bacterial species. Sequencing of the mycoplasma PCR products revealed a close genetic relationship to <i>Mycoplasmopsis agassizii</i>. TGS identified genetically distinct mycoplasmas and three different serpentoviruses. While mycoplasmas could not be successfully propagated, <i>Brucella intermedia</i> comb. nov. was identified in eight cultures. Respiratory disease in snakes is often multifactorial, involving various pathogens and environmental influences. This study demonstrates that comprehensive diagnostics are essential for understanding disease processes in snakes and improving the detection of diverse pathogens. Further research is needed to improve laboratory diagnostics for infectious diseases in reptiles and to better understand the roles of various pathogens in respiratory diseases in snakes.

Abstract (opening): The commercial wildlife trade involves billions of animals each year, consumed for various purposes, including food, fashion, entertainment, traditional medicine, and pets. The experiences of the animals involved vary widely, with negative welfare states being commonplace. To highlight the broad scope of animal welfare impacts across the commercial wildlife trade, we present ten case studies featuring a range of species traded globally for different purposes: (1) Ball pythons captured and farmed to serve as pets; (2) Zebrafish captive bred to serve as pets; (3) African Grey Parrots taken from the wild for the pet industry; (4) Sharks de-finned for traditional medicine; (5) Pangolins hunted for traditional medicine; (6) Crickets farmed for food and feed; (7) Frogs wild-caught for the frog-leg trade; (8) Crocodilians killed for their skins; (9) Lions farmed and killed for tourism; and (10) Elephants held captive for tourism. The case studies demonstrate that wild animals commercially traded can suffer from negative welfare states ranging from chronic stress and depression to frustration and extreme hunger. The individuals involved range from hundreds to billions, and their suffering can last a lifetime. Given the welfare issues identified and the growing recognition and scientific evidence for animal sentience, we propose reducing and redirecting consumer demand for these consumptive wildlife practices that negatively impact animals.

Abstract (opening): Dystocia represents a multifactorial and clinically significant reproductive disorder affecting a broad spectrum of reptilian species. Commonly resulting from prolonged vitellogenesis, endocrine disruption, or hepatic lipidosis, dystocia is often exacerbated by suboptimal husbandry or concurrent disease. This review critically evaluates the etiology, diagnostic criteria, and therapeutic interventions associated with this condition. Emphasis is placed on the interplay between metabolic exhaustion and hepatic compromise, which may lower the threshold for surgical intervention. The efficacy and limitations of oxytocin-based protocols are discussed in the context of hormonal receptor variability and response attenuation. Advanced diagnostic modalities, including ultrasonography, radiography/CT, and biochemical profiling, are reviewed for their utility in case stratification. Finally, surgical management options are considered for cases refractory to medical treatment, with attention paid to timing, anesthetic risk, and post-operative care. Collectively, this synthesis aims to inform evidence-based clinical decision-making and promote improved standards of care in reptile reproductive medicine.

Abstract (opening): <h4>Background</h4>Respiratory diseases are among the main causes of morbidity and mortality in captive reptiles. In Romania, pneumonia is a frequently observed illness affecting pet reptiles. Key factors contributing to the high incidence of pneumonia include inadequate animal husbandry, poor nutrition, and insufficient hygiene practices. Bacteria may act as primary pathogens or as facilitators of disease severity.<h4>Methods</h4>This study investigates bacterial strains from multiple genera and species (<i>Chryseobacterium</i> (<i>C.</i>) <i>indologenes</i>, <i>Staphylococcus</i> (<i>S.</i>) <i>epidermidis</i>, <i>Escherichia</i> (<i>E.</i>) <i>coli</i>, and <i>Pseudomonas</i> (<i>P.</i>) <i>aeruginoasa</i>) from six ball pythons regarding their antibiotic susceptibility and the effect of essential oils. Bacteria were isolated from the lower respiratory tract, displaying clinical signs of pneumonia. All isolates were tested with essential oils (lemongrass, oregano, rosemary, and sage) and a grapefruit seed extract (GSE) at different dilutions.<h4>Results</h4>The incidence of <i>Chryseobacterium indologenes</i> was highest (3 isolates/12 samples, 25%), followed by <i>E. coli</i> and <i>Staphylococcus epidermidis</i> (2/12 each, 16.6%), and <i>Pseudomonas aeruginoasa</i> (1/12, 8.3%). Resistance profiling to different antibiotic classes revealed that all isolates (eight) were resistant to multiple antibiotics tested by us. All isolates were resistant to β-lactams and fluoroquinolones. One strain of <i>E. coli</i> exhibited intermediate resistance to quinolone and penicillin. All strains were categorized as multidrug-resistant. GSE showed antibacterial activity against all isolates.<h4>Conclusions</h4>Wanting to deepen our understanding of the microorganisms that can infect ball pythons and recognizing that all isolated bacteria have zoonotic potential, this paper highlights some common issues faced by exotic animal owners and suggests that treatments should also include the use of essential oils.

Abstract (opening): Animal models enable investigation of the impact of stress on emotional-behavioral performance to facilitate understanding of posttraumatic stress symptoms experienced in humans. Refinement of animal stress models could lead to a reduction in the number of subjects needed to detect statistically significant stress effects, in accordance with Russel and Burch's three Rs of research. We assessed whether performance of experimental procedures (that is, stress exposure and poststress behavioral testing) during the dark or light phases of the 12-h light/12 h-dark cycle is a refinement that could accomplish this reduction. At 3 h into either the light or dark phase, male and female adult Sprague-Dawley rats underwent a single-day traumatic stress exposure. Rats then underwent behavioral testing for exploratory behaviors, startle responses, and conditioned fear responses at 2 h, 1 d, and 9 d after stress exposure. Distance traveled in the elevated plus maze (EPM) by both male and female rats was significantly reduced in the dark phase compared with the light phase. Male rats of the dark phase group also spent less time in the open arms of the EPM, and traveled less, spent less time in the center, and spent more cumulative time freezing in the open field. Female rats of the dark phase group spent more cumulative time freezing in the EPM and exhibited significantly more tone-cued conditioned freezing responses. Our results suggest that performing experimental procedures during the dark phase of the light cycle may be a useful refinement mechanism, as procedures performed during this period had the greatest effect on behavioral outcomes in both males and females. Light cycle phase is an experimental variable that should be considered when designing experiments to maximize behavioral effects, including those in response to stress.

Abstract (opening): Iguanas exhibit diverse colors and behaviors reflecting evolutionarily adaptation to various habitats; in particular, the Galápagos iguanas represent unique color morphologies with distinct ecological niches. While external coloration in iguanas has ecological implications, comprehensive studies on the histological and ultrastructural aspects of their skin can provide insight into their adaptation to extreme environments, such as high UV exposure. Starting from these considerations the present study investigates the histological, ultrastructural and immunohistochemical features to comprehensively characterize the skin in adults of three species of Galápagos iguanas (A. cristatus, C. subcristatus and C. marthae). Morphological analysis revealed significant differences among the species, with the black-colored skin of A. cristatus showing a melanin-rich but vessel-poor dermis, while C. subcristatus and C. marthae displayed varying distributions of melanosomes and melanocytes. Notably, the absence of iridophores was consistent across all samples due to the absence of birefringent material under the optical microscope. Morphometric evaluations highlighted interspecific differences in the stratum corneum thickness, particularly between black- and non-black-colored (irrespectively if yellowish or pink) skin. The ultrastructural investigation confirmed the absence of iridophores in all analyzed samples. The cytokeratin expression assessed by immunohistochemistry showed stratified epithelium in the epidermis of C. marthae non-black-colored (pink) skin. The presence of a thickened stratum corneum and the stratification of the epidermis in non-pigmented skin could help the pink iguana to cope with the extreme conditions of the Wolf volcano, especially in relation to UV exposure. These skin characteristics may reduce the penetration power of UV rays into the superficial loose dermis, thereby attenuating potential UV-related damage such as DNA breaks and ROS generation. These findings offer insights into the adaptive strategies of these iguanas.

Abstract (opening): Given the growing number of events involving exotic animals, it is crucial to prioritize the well-being of the animals involved. This study aims to evaluate the quality of animal presentation at a selected fair in Poland and assess the level of animal welfare evident in the exhibition boxes, contributing to the ongoing dialogue on this important issue. The evaluators used a five-point Likert scale and a Yes/No system to analyze the living conditions during the fair, including the size of containers, presence of substrate, and environmental enrichment. They also assessed the occurrence of visual abnormal postures and behaviors to gauge the overall level of welfare. To ensure the reliability and consistency of the data and minimize potential bias, each evaluator repeated the rating process three times, with a three-week interval between each session. An average value was then calculated for each aspect. A total of 818 animals were present at the fair, with 688 being reptiles (84.11%) and 130 being amphibians (15.89%). This study revealed that the provision of substrate scored higher for reptiles compared to amphibians, while the size of containers for amphibians received higher ratings than those for reptiles. Visual abnormalities in posture and behavior were more common in reptiles than in amphibians. Display containers for snakes received the lowest ratings and showed more visual abnormalities in posture and behavior, raising concerns about their welfare. Despite the presence of environmental enrichment, the overall level of animal welfare was assessed as being medium/low. Pearson's correlation coefficient indicated good reliability among the evaluators during the assessment process, with most assessments showing values > 0.8. Despite existing regulations for exhibitors, neglect remains prevalent. These findings highlight the potential negative impact of animal exposure at fairs on animal welfare. Display containers were often inadequately sized for the animals, particularly for snakes, chameleons, monitor lizards, and salamanders.

Abstract (opening): There is an ethical need to document and develop best practices for meeting ambassador animals' welfare needs within the context of meeting zoo and aquarium program objectives. This is because ambassador animals experience direct and frequent contact with humans. This paper rigorously synthesizes behavioral research and theory, contemporary practices, and personal experiences to offer key concepts that can be applied to meet ambassador animal welfare needs. These key concepts include addressing an animal's recognition of choice and control, the use of the most positive and least intrusive effective interventions when training animals to participate in programming, and an overall reduction in aversive strategy use. Our model for increasing ambassador animal welfare focuses on seven main areas of concern, including the following: choosing the most suitable animal for the program; choosing the human with the right skills and knowledge for the program; using the most positive, least intrusive, effective training methods; developing a strong trusting relationship between trainer and animal; developing a comprehensive enrichment program; the need for institutional support; and creating opportunities for animals to practice species-appropriate behaviors. Our model will provide guidelines for improved ambassador animal welfare that can be refined with future research.
